Duchenne muscular dystrophy (DMD) is the most common muscle disease in childhood and affects almost exclusively boys. It is caused by a genetic mutation that leads to absent or faulty production of the muscle protein dystrophin. Motor impairments become apparent in early childhood, and by around age 10, most affected individuals require a wheelchair. Common signs include delayed walking, unusually large calf muscles, and a waddling gait. Although DMD is currently incurable, modern therapies—including corticosteroids, physiotherapy, and new genetic approaches—can slow disease progression and improve quality of life. New medications and ongoing gene therapy trials offer hope for the future.
